چکیده

p73, a recently identified gene highly homologous to p53, can transactivate p53 target genes and induce apoptosis. Here we report the identification of an NH2-terminal truncated isoform of human p73, Np73, which is capable of suppressing p53and p73-dependent transactivation. We speculate that this suppression is achieved by competing for the DNA binding site in the case of p53 and by direct association in the case of TAp73. Expression of Np73 in cancer cell lines also inhibited suppressive activity of p53 and TAp73 in colony formation, implying possible involvement of Np73 in oncogenesis by inhibiting the tumor-suppressive function of p53 and TAp73. Also reported is the identification of TAp73 , a new member of the COOH-terminal truncated isoform of p73 and tissuespecific expression of these isoforms, along with other previously identified p73 isoforms.

∆Np73 is capable of inducing apoptosis by co-ordinately activating several BH3-only proteins

Inactivation of p53 is one of the most relevant events in human cancer, since it allows transformed cells to escape their own proliferation control and leave them irresponsive to drugs that aim to damage their DNA. When p53 falls, other members of its family may become targets to attack tumoural cells. p73 has shown capacity to mediate these attacks. However, its N-terminal truncated isoforms h...

Advances in Brief p73 Isoforms Can Induce T-Cell Factor–Dependent Transcription in Gastrointestinal Cells

A new p53 family member, p73, and its isoform Np73 are increasingly recognized in cancer research as important players in tumorigenesis, as well as in chemotherapeutic drug sensitivity. Despite substantial structural similarities to p53, accumulating evidence suggests that p53 and p73 may play different roles in human tumorigenesis. In this study, we have investigated the role of p73 and Np73 i...

Role of p73 Dinucleotide Polymorphism in Prostate Cancer and p73 Protein Isoform Balance

Background. Molecular markers for prostate cancer (PCa) risks are currently lacking. Here we address the potential association of a dinucleotide polymorphism (DNP) in exon 2 of the p73 gene with PCa risk/progression and discern any disruption of p73 protein isoforms levels in cells harboring a p73 DNP allele. Methods. We investigated the association between p73 DNP genotype and PCa risk/aggress...

Expression of Np73 Predicts Poor Prognosis in Lung Cancer

Purpose: Np73 is an isoform of the p53 homologue p73, which lacks an NH2-terminal transactivation domain and antagonizes the induction of gene expression by p53/ p73. The aim of this study was to detect Np73 expression in lung cancer and to evaluate the relationship between the Np73 expression level and the prognosis of patients with resected lung cancer. Experimental Design: We used immunohist...
